Phagosomal signaling by Borrelia burgdorferi in human monocytes involves Toll-like receptor (TLR) 2 and TLR8 cooperativity and TLR8-mediated induction of IFN-beta [Immunology]

Phagocytosed Borrelia burgdorferi (Bb) induces inflammatory signals that differ both quantitatively and qualitatively from those generated by spirochetal lipoproteins interacting with Toll-like receptor (TLR) 1/2 on the surface of human monocytes. Of particular significance, and in contrast to lipoproteins, internalized spirochetes induce transcription of IFN-β. Using inhibitory immunoregulatory DNA sequences (IRSs) specific to TLR7, TLR8, and TLR9, we exhibit that the TLR8 inhibitor IRS957 significantly diminishes production of TNF-α, IL-6, and IL-10 and completely abrogates transcription of IFN-β in Bb-stimulated monocytes. We demonstrate that live Bb induces transcription of TLR2 and TLR8, whereas IRS957 interferes with their transcriptional regulation. Using …

Exhaled nitric oxide and spirometry in respiratory health surveillance

Conclusions FENO and spirometry were not overlapping dimensions in ABs and hairdressers, each check contributing unique data on the physiological status of the respiratory system. FENO may provide added data on airway inflammation not provided by spirometry. Cofilin-1 in the podocyte: a molecular switch for actin dynamics

Summary of key findings Studies by Garg et al. and Ashworth et al. investigated the functional relevance of a key regulatory protein, cofilin-1, for
podocyte actin dynamics (Ashworth et al. in PLos One 5:e12626, 2010; Garg et al. in J Biol Chem 285:22676–22688, 2010). Using
different imitation organisms (zebrafish or transgenic mice), both groups observed a collapse of the glomerular filtration barrier
upon inactivation of cofilin-1. In elegant biochemical studies, Garg et al. established that cofilin-1 activity is regulated
by nephrin, which is part of the slit diaphragm complex. Two feedback loops stabilize cofilin-1 in the phosphorylated versus
dephosphorylated state. The …

Asthma Through The Eyes Of A Medical Anthropologist

Asthma affects an estimated 8 percent of Americans, and about 300 million human beings encircling the earth, however varying practices in diagnosis and treatment have global implications in understanding a widespread, chronic condition, says Van Sickle, who applies an anthropological approach to medicine. "Since the major path to learn how many human beings have asthma is to inquiry them, external factors that alter those estimates have a major impact on our understanding of how widespread asthma is," he says. "Yet community culture and conditions constitute these estimates subject to a fantastic deal of mistake... (Source: Health News from Medical News …

The role of pseudoephedrine on daytime somnolence in patients suffering from perennial allergic rhinitis (PAR).

Authors: Sherkat AA, Sardana N, Safaee S, Lehman EB, Craig TJ
Allergic rhinitis is one of distinct inflammatory diseases affecting the nasal mucosa. Cellular inflammation of nasal mucosa is a hallmark of this disease and is characterized by the accumulation of eosinophils and the release of various chemical messengers such as chemokines, cytokines, and histamine. This inflammation of the nose leads to nasal congestion and a reduction in sleep quality, resulting in daytime somnolence.
